Ryan Bowman failed to recover from illness and was unable to take part for Hereford on their first visit to Kenilworth Road for almost 37 years.
Injured Marley Watkins also failed to make the trip which saw United revert to a 4-3-1-2 formation with Sam Clucas in the hole behind strike pairing Marlon Jackson and Chris Sharp.
Joe Connor reverted to right-back with Chris Carruthers getting his first start in five months in a midfield role.
Former Bulls Janos Kovacs, who was injured, and Stuart Fleetwood, who was not, failed even to make the bench for the home side.
Luton made the early running and a shot from Alex Lawless flashed across goal and out for a corner.
Lawless took the kick but James Bittner made a good catch.
Hereford hit back and a deflected cross from Clucas almost fell to Jackson.
Caruthers was then brought down on the edge of the area and Jackson’s free-kick deflected behind for a corner.
From Clucas’ cross, Stefan Stam saw a  header cleared off the line.
Another glorious chance went begging on nine minutes when Clucas got around the back of the home defence and Josh O’Keefe’s shot, heading for a seemingly empty net, was deflected out for a corner off a defender.
Sharp tried his luck after good work by Jackson but his shot was weak and easily fielded by goalkeeper Mark Tyler.
Sharp blazed high over again on 14 minutes after good work by O’Keefe and Clucas.
The Luton attack made its presence felt on 19 minutes with good work by Jake Howells whose 20-yarder drifted wide.
On 23 minutes, Hereford took the lead with an excellent goal.
Jackson received the ball in midfield and used his pace to ease pace two defenders before sliding a shot past Tyler.
Luton enjoyed the better of the next passage of play and Arnaud Mendy drilled in a powerful shot which flew straight into the arms of Bittner.
Andre Gray, Town’s most dangerous forward, got to the bye-line but his cross was over the heads of all his team-mates.
Howells also let fly from 30 yards but straight at Bittner.
Jackson, who was looking a real threat each time he gets on the ball, weaved into the area but his shot was deflected behind for a corner.
Luton won a corner in stoppage time but Bittner caught easily.
As Hereford attacked at the start of the second period, Sharp’s shot took a deflection for a corner.
There was a let-off for Hereford, though, with a Jonathan Smith shot which hit the bar, a post and then was cleared for a corner.
Luton were beginning to have the better of play and Howells found a lot of space on the left of the area but thrashed his shot past the far post.
Connor conceded another corner on 70 minutes but Stam was again alert to the danger and get s the ball over the bar. 
Luton continued to press and, after some unexpected trickery from the tall Alex Wall on the left, Gray side-footed narrowly over.
Hereford staged a rare attack and Jackson clipped a neat ball in to Clucas who scuffed his shot to allow an easy save.
But the Bulls almost got a bizarre second on 80 minutes when David Martin, in attempting to clear, fired the ball against his own bar.
Martin had better luck at the other end, three minutes from time, firing in a left-footer from the edge of the area.
